Anyone who done this, can I drive from daan/Taipei to Kaohsiung in one day ? I’m on a red plate motorcycle so I can use express way but not freeway
How long would it take ? Also, what route is recommended ?
You can do it, you have plenty of (Taiwanese) roads. If time is an issue, you can take expressways 15 and 61, they are on and off (or maybe they are finally totally constructed?), but they are the fastest way to go from one tip of the island to the other on two wheels.

64 from Taipei to Bali (or just take the bridge if you’re in North Taipei), 61 from Bali to Tainan, then 17 to Kaohsiung. Around 6 hours.
